Therefore, it is only possible to connect to on-premise databases by adding the IP addresses on the firewall whitelist. However, this connection is through the Cloud Database option (even if it is on-prem), so can this method be used in Enterprise Standard licenses or is it necessary to upgrade to Plus?

Enterprise Standard can connect to a database with a public IP address. Is that what you're asking?

Yes! It needs to be public or can we just add the Google/AppSheet IP addresses to the allowlist?

The database must have a public IP address, not a private address. Absolutely you should protect the database with an allowlist!
